Oxidative bisulphite sequencing allows differentiation between 5-methylcytosine and 5-hydroxymethylcytosine in genomic DNA.

In this method, deep sequencing of oxidative bisulphite-treated DNA and sequence comparison of treated vs untreated libraries can quantify and map 5-hydroxymethylated locations at single-base resolution, and at genome wide scale.
